    Abstract:

Applications: The Invention provides a thermostable phenylalanine conjugated human insulin derivative comprising an extra phenylalanine residue conjugated through an isopeptide bond between the side chain amine of Lysine B29 of human insulin and carboxyl terminal of introduced phenylalanine.
Advantages: 1. The method has technically advantageous part that this is the highly simple one step modification procedure to obtain a thermostable human insulin variant. This modification does not incorporate any non-natural or halogenated amino acids to the native human insulin. 2. Also, the method does not require any exclusive, time consuming as well as low yielding total synthesis or multistep conjugation or solid phase synthesis protocol which makes it highly cost effective. 3. Underprivileged patients need proper solutions to thermal degradation issue to enjoy insulin usage without any stability drawback. 4. Reducing the thermolabile property of insulin would be helpful for its longer storage in diverse unfavourable conditions. 5. There is a need of stabilizing techniques to maintain insulin viability for longer duration without costly cold chain facilities. 6. Commercial reoptimization is also not required for the major industrial plants for insulin overexpression and purification steps. 7. To find out the stability of this newly designed insulin analogue even under lower pH condition, ThT fluorescence was carried out in acidic condition.
