The protein AMP activated Protein Kinase (AMPK) is an energy stress sensor that is activated under conditions that lead to a reduction in energy, such as under hypoxia and nutrient deprivation. Upon activation AMPK acts as a central metabolic regulator to bring about energy homeostatis by inhibiting energy consuming anabolic activities, while activating energy producing catabolic activities. Owing largely to its antigrowth effects, it earned its name as tumor-suppressor. In contrast, our findings identify novel protumorigenic functions of AMPK, particularly in tumor progression leading to metastasis. For eg, we have shown that AMPK is critical for cancer cells to survive matrix detachment, as well as in promoting epithelial-mesenchymal transition (EMT) two key processes that contribute to metastasis (spread of tumor). Thus, we have identified AMPK activation, and thus metabolic alteration, as a key switch that promotes breast cancer progression. Based on our findings, we advocate AMPK inhibition as novel strategy to reduce the burden of metastatis . The major reason for cancer associated deaths. Currently there are no FDA approved AMPK inhibitors available for use in the clinic. Hence, identifying novel AMPK inhibitors, and testing their effect in preventing cancer spread, can have therapeutic application.
