We are screening FDA approved drugs against Non-structural protein 1 (Nsp1) of SARS-CoV-2. By binding to the 40S ribosomal subunit of host cells, Nsp1 renders it translationally inactive. Nsp1 helps the virus in evading the host immune system by suppressing the production of interferons thereby affecting the entire host anti-viral response. Hence Nsp1 is an attractive antiviral target.

    Abstract:

Applications: The C-terminal helices of Nsp1 binds in the mRNA channel of the 40S and hence blocks protein synthesis in the host. This helps the virus to escape the host immune system as the production of interferons is suppressed. Hence, the anti-viral response by the host is blocked. Therefore, the combinational approach to figure out if any currently available FDA approved drugs can bind to the C-terminal helices of Nsp1 and hence that would prevent binding to 40S. Thus, such drugs can be repurposed for Covid-19.
Advantages: The drugs that would be shortlisted in this study will be FDA approved, hence, such drugs can be immediately repurposed for Covid-19.
