Description : The High-performance liquid chromatography (analytical & preparative) was installed in the Department of Microbiology, Periyar University, Salem-11. High-performance liquid chromatography or commonly known as HPLC, is an analytical technique used to separate, identify or quantify each component in a mixture. The HPLC (Shimizu Japan) is equipped with photodiode array detector and florescent detector for analysis and separation of organic molecules. The preparative (C18, 10µm, 20 ×250mm) and analytical (C18, 5µm, 4.6 ×250mm) column used for HPLC analysis. The column packing is non-polar; the mobile phase is water+ miscible solvent (e.g Methanol and acetonitrile). It can be used for polar, non-polar, ignitable, and ionic samples. The total solvent flow rate is 1.0 ml/min analytical and 10.0 ml/min preparative. The column temperature will be maintained at 40? and pressure is set as maximum 449mpa.
